RESUMO

Two experiments were conducted to investigate the production effects of N-acetyl-l-methionine (NALM; experiment 1) and to estimate its bioavailability (BA) and rumen escape (RE; experiment 2), respectively, in lactating dairy cows. In experiment 1, 18 multiparous Holstein cows were used in a replicated, 3 × 3 Latin square design experiment with three 28-d periods. Treatments were (1) basal diet estimated to supply 45 g/d digestible Met (dMet) or 1.47% of metabolizable protein (MP; control), (2) basal diet top-dressed with 32 g/d of NALM to achieve dMet supply of 2.2% of MP, and (3) basal diet top-dressed with 56 g/d of NALM to achieve dMet supply of 2.6% of MP. The NALM treatments supplied estimated 17 and 29 g/d dMet from NALM, respectively, based on manufacturer's specifications. In experiment 2, 4 rumen-cannulated lactating Holstein cows were used in a 4 × 4 Latin square design experiment with four 12-d periods. A 12-d period for baseline data collection and 4 d for determination of RE of NALM preceded the Latin square experiment. For determination of RE, 30 g of NALM were dosed into the rumen simultaneously with Cr-EDTA (used as a rumen fluid kinetics marker) and samples of ruminal contents were collected at 0 (before dosing), 1, 2, 4, 6, 8, 10, 14, 18, and 24 h after dosing. Rumen escape of NALM was calculated using the estimated passage rate based on the measured Cr rate of disappearance. Bioavailability of abomasally dosed NALM was determined using the area under the curve of plasma Met concentration technique. Two doses of l-Met (providing 7.5 and 15 g of dMet) and 2 doses of NALM (11.2 and 14.4 g of dMet) were separately pulse-dosed into the abomasum of the cows and blood was collected from the jugular vein for Met concentration analysis at 0 (before dosing), 1, 2, 4, 6, 8, 10, 12, 14, 18, and 24 h after dosing. Supplementation of NALM did not affect DMI, milk yield, feed efficiency, or milk protein and lactose concentrations and yields in experiment 1. Milk fat concentration and energy-corrected milk yield decreased linearly with NALM dose. Plasma Met concentration was not affected by NALM dose. The estimated relative BA of abomasally dosed NALM (experiment 2) was 50% when dosed at 14.4 g/cow (11.2 g/d dMet from NALM) and 24% when dosed at 28.8 g/cow (14.4 g/d dMet from NALM). The estimated RE of NALM was 19% based on the measured kp of Cr at 11%/h. The total availability of ingested NALM was estimated at 9.5% for the lower NALM dose when taking into account RE (19%) and bioavailability in the small intestine (50%). Overall, NALM supplementation to mid-lactation dairy cows fed a MP-adequate basal diet below NRC (2001) recommendations (45 g/d or 1.47% Met of MP) decreased milk fat and energy-corrected milk yields but did not affect milk or milk true protein yields. Further evaluation of BA of NALM at different doses is warranted. In addition, intestinal conversion of NALM to Met needs additional investigation to establish a possible saturation of the enzyme aminoacylase I at higher NALM doses.

The objective of this experiment was to evaluate the effects of supplementing a rumen-protected source of Met, N-acetyl-l-methionine (NALM), on lactational performance and nitrogen metabolism in early- to mid-lactation dairy cows. Sixty multiparous Holstein dairy cows in early lactation (27 ± 4.3 d in milk, SD) were assigned to 4 treatments in a randomized complete block design. Cows were blocked by actual milk yield. Treatments were as follows: (1) no NALM (control); (2) 15 g/d of NALM (NALM15); (3) 30 g/d of NALM (NALM30); and (4) 45 g/d of NALM (NALM45). Diets were formulated using a Cornell Net Carbohydrate and Protein System (CNCPS) v.6.5 model software to meet or exceed nutritional requirements of lactating dairy cows producing 42 kg/d of milk and to undersupply metabolizable Met (control) or supply incremental amounts of NALM. The digestible Met (dMet) supply for control, NALM15, NALM30, and NALM45 were 54.7, 59.8, 64.7, and 72.2 g/d, respectively. The supply of dMet was 88, 94, 104, and 115% of dMet requirement for control, NALM15, NALM30, and NALM45, respectively. Milk yield data were collected, dry matter intake (DMI) was measured daily, and milk samples were collected twice per week for 22 wk. Blood, ruminal fluid, urine, and fecal samples were collected during the covariate period and during wk 4, 8, and 16. Data were analyzed using the GLIMMIX procedure of SAS (SAS Institute) using covariates in the model for all variables except body weight. Linear, quadratic, and cubic contrasts were also tested. Treatments did not affect DMI, milk yield, and milk component concentration and yield; however, feed efficiency expressed as milk yield per DMI and 3.5% fat-corrected milk per DMI were quadratically affected, with greater response observed for NALM15 and NALM30 compared with control. Acetate proportion linearly increased, whereas propionate proportion linearly decreased with NALM supplementation. Blood urea nitrogen linearly decreased with NALM supplementation. Total plasma essential AA concentrations were quadratically affected, as greater values were observed for control and NALM45 than other treatments. Plasma Met concentration was quadratically affected as lower levels were observed with NALM15, whereas Met concentrations increased with NALM45 compared with control. Nitrogen utilization efficiency and apparent total-tract nutrient digestibility were not affected by treatment. Supplementation of NALM at 15 or 30 g/head per day resulted in the greatest improvements in feed efficiency without affecting N metabolism of early- to mid-lactation dairy cows.

The present study investigated production responses and ruminal fermentation characteristics of lactating dairy cows when supplemented with N-acetyl-l-Met (NALM) as a source of rumen-protected Met in metabolizable protein (MP)-deficient (MPD) or MP-adequate diet (MPA). Eight lactating dairy cows (53 ± 10.4 d in milk, average ± standard deviation) were blocked by parity and days in milk, and the experiment was performed in a replicated 4 × 4 Latin square design. Within each square, cows were randomly assigned to a sequence of 4 diets during each of the four 21-d periods (14 d of treatment adaptation and 7 d of data collection and sampling). A 2 × 2 factorial arrangement was used; MPD or MPA was combined without or with NALM: MPD without NALM, MPD with NALM (MPD+NALM), MPA without NALM, and MPA with NALM (MPA+NALM). A NALM product was supplemented in the MPD+NALM and the MPA+NALM at 30 g/cow per d. Supplementation of NALM did not affect dry matter intake (DMI) and milk yield regardless of MP concentration. In addition, supplementing NALM resulted in a similar milk true protein concentration and yield. In contrast, NALM supplementation increased milk fat concentration and yield and 3.5% fat-corrected milk yield and tended to increase energy-corrected milk yield regardless of MP difference. Additionally, trends were observed for increased 3.5% fat-corrected milk yield/DMI and energy-corrected milk yield/DMI, and the positive effects were greater under the MPA than the MPD diet, resulting in trends toward interactions between MP and NALM. Dietary treatments had similar effects on ruminal fermentation characteristics and microbial protein yield. Plasma concentration of Met increased under the MPD but not the MPA diet, leading to an MP × NALM interaction. Overall results in the current study suggest that NALM exerted a minor influence on ruminal metabolism, but increased milk fat concentration, resulting in increases in milk fat yield and feed efficiency. Yet, potential effects of NALM on intermediary metabolism between the gastrointestinal tract, the liver, and the mammary gland need to be explored to understand utilization efficiency for production of dairy cows.

This experiment was conducted to determine effects of feeding birdsfoot trefoil hay-based diets in comparison with an alfalfa hay-based diet on N utilization efficiency, ruminal fermentation, and lactational performance by mid-lactation dairy cows. Nine multiparous lactating Holstein cows (131 ± 22.6 d in milk), 3 of which were rumen fistulated, were fed 3 experimental diets in a replicated 3 × 3 Latin square design with 3 periods of 14 d of adaptation and 7 d of data and sample collection. Within squares, cows were randomly assigned to diets as follows: alfalfa hay-based diet (AHT), alfalfa and birdsfoot trefoil hay-based diet (ABT), and birdsfoot trefoil hay-based diet (BT). Intakes of dry matter and crude protein were similar across treatments, whereas ABT and BT diets resulted in decreased fiber intake compared with AHT. Feeding BT tended to increase neutral detergent fiber digestibility compared with AHT and ABT. Milk yield tended to increase for cows consuming ABT or BT diets. Milk true protein concentration and yield were greater for cows consuming ABT relative to those fed AHT. Concentration of total volatile fatty acids tended to increase by cows fed BT compared with those fed AHT and ABT. Feeding birdsfoot trefoil hay in a total mixed ration resulted in a tendency to decrease acetate proportion, but it tended to increase propionate proportion, leading to a tendency to decrease acetate-to-propionate ratio. Whereas concentration of ammonia-N was similar across treatments, cows offered BT exhibited greater microbial protein yield relative to those fed AHT and ABT. Cows offered birdsfoot trefoil hay diets secreted more milk N than AHT, resulting in improved N utilization efficiency for milk N. The positive effects due to feeding birdsfoot trefoil hay were attributed to enhanced neutral detergent fiber digestion, and thus it could replace alfalfa hay in high-forage dairy diets while improving N utilization efficiencies and maintaining lactational performance compared with alfalfa hay.

This study was carried out to investigate the effects of intraruminal infusion of propionate on ruminal fermentation characteristics and blood hormones and metabolites in Hanwoo (Korean cattle) steers. Four Hanwoo steers (average body wt. 270 kg, 13 month of age) equipped with rumen cannula were infused into rumens with 0.0 M (Water, C), 0.5 M (37 g/L, T1), 1.0 M (74 g/L, T2) and 1.5 M (111 g/L, T3) of propionate for 1 hour per day and allotted by 4×4 Latin square design. On the 5th day of infusion, samples of rumen and blood were collected at 0, 60, 120, 180, and 300 min after intraruminal infusion of propionate. The concentrations of serum glucose and plasma glucagon were not affected (p>0.05) by intraruminal infusion of propionate. The serum insulin concentration at 60 min after infusion was significantly (p<0.05) higher in T3 than in C, while the concentration of non-esterified fatty acid (NEFA) at 60 and 180 min after infusion was significantly (p<0.05) lower in the propionate treatments than in C. Hence, intraruminal infusion of propionate stimulates the secretion of insulin, and decreases serum NEFA concentration rather than the change of serum glucose concentration.

AIMS: To identify whether the supplement of anaerobic fungi isolates with cellulolytic activities accelerates the silage fermentation. METHODS AND RESULTS: Three fungal isolates with the highest cellulolytic activities among 45 strains of anaerobic fungal stock in our laboratory were selected and used as silage inoculants. The rice straw (RS) was ensiled for 10, 30, 60, 90 and 120 days with four treatments of anaerobic fungi derived from the control (no fungus), Piromyces M014 (isolated from the rumen of the Korean native goat), Orpinomyces R001 (isolated from the duodenum of Korean native cattle) and Neocallimastix M010 (isolated from the guts of termites), respectively. The silages inoculated with pure strains of fungi showed a higher fungal population (P < 0.05) when compared to the control silage. In situ ruminal DM disappearance of RS silage (RSS) was improved with fungal treatment. SEM observation showed live fungal cells inoculated in RS could survive during the ensiling process. Overall, this study indicated that the inoculation of anaerobic fungi decreased the cell wall content of the RSS and increased in situ dry matter disappearance. CONCLUSIONS: The supplementation of anaerobic fungi isolates to RSS as a silage inoculant improves the RSS quality. SIGNIFICANCE AND IMPACT OF THE STUDY: This is the first study showing the potential application of supplement of anaerobic fungi isolated from the guts may be applied industrially as an alternate feed additive that improves the silage quality.

This experiment was performed to test a hypothesis that nutritive benefits of feeding high-moisture corn (HMC) would be different when fed with different qualities of alfalfa hay (AH) due to associative effects on ruminal fermentation and nutrient utilization efficiency. Eight multiparous lactating Holstein cows were used; 4 were surgically fitted with ruminal cannulas. Days in milk averaged 184 ± 10.7 at the start of the experiment. The experiment was performed in a duplicate 4 × 4 Latin square design. Within each square, cows were randomly assigned to a sequence of 4 diets during each of the four 21-d periods (14 d of treatment adaptation and 7 d of data collection and sampling). A 2 × 2 factorial arrangement was used; fair-quality AH [FAH; 39.6% neutral detergent fiber (NDF) and 17.9% crude protein (CP)] or high-quality AH (HAH; 33.6% NDF and 21.9% CP) was combined with steam-flaked corn (SFC) or HMC to form 4 treatments: FAH with SFC, FAH with HMC, HAH with SFC, and HAH with HMC. The AH was fed at 32% dry matter (DM) content, whereas SFC or HMC was included at 17% DM content. Quality of AH did not affect DM intake, whereas feeding HMC decreased DM intake, regardless of quality of AH. Digestibility of DM was greater for cows fed HAH compared with those fed FAH (70.1 vs. 67.6%). Digestibility of NDF increased by feeding HMC (67.6 vs. 58.4%), but not by quality of AH. Under FAH, starch digestibility decreased by feeding HMC compared with SFC (85.7 vs. 95.0%), but it was similar under HAH, resulting in an interaction between quality of AH and type of corn grain (CG). Feeding different qualities of AH did not affect milk yield; however, feeding HMC decreased milk yield in FAH diet, causing an AH × CG interaction. Efficiency of milk yield/DM intake was improved due to feeding HMC, regardless of the quality of the AH. In addition, dietary N utilization for milk N tended to increase by feeding HMC, but it was not influenced by quality of AH. Yield of microbial protein increased by feeding HAH diets compared with FAH diets, whereas feeding the HMC diet increased microbial protein yield under the HAH diet, leading to an interaction between and AH and CG. Overall results in this experiment indicate that feeding HMC in AH-based diets improved feed efficiency as well as N utilization efficiency, regardless of quality of AH.

Due to the increasing cost of soybean meal and concerns of excess N being excreted into the environment, new protein supplements have been developed. Two products that have shown potential in increasing N utilization efficiency are slow-release urea (SRU; Optigen; Alltech Inc., Nicholasville, KY) and ruminal-escape protein derived from yeast (YMP; DEMP; Alltech Inc.). The objective of this study was to assess the effects of feeding these 2 supplements in alfalfa hay-based [45.7% of forage dietary dry matter (DM)] dairy diets on nutrient utilization, feed efficiency, and lactational performance of dairy cows. Twelve multiparous dairy cows were used in a triple 4 × 4 Latin square design with one square consisting of ruminally cannulated cows. Treatments included (1) control, (2) SRU-supplemented total mixed ration (SRUT), (3) YMP-supplemented total mixed ration (YMPT), and (4) SRU- and YMP-supplemented total mixed ration (SYT). The control consisted only of a mixture of soybean meal and canola meal in a 50:50 ratio. The SRU and the YMP were supplemented at 0.49 and 1.15% DM, respectively. The experiment consisted of 4 periods lasting 28 d each (21 d of adaptation and 7 d of sampling). Cows fed YMPT and SYT had decreased intake of DM, and all supplemented treatments had lower crude protein intake compared with those fed the control. Milk yield tended to have the greatest increase in YMPT compared with the control (41.1 vs. 39.7 kg/d) as well as a tendency for increased milk fat and protein yields. Feed efficiencies based on yields of milk, 3.5% fat-corrected milk, and energy-corrected milk increased at 10 to 16% due to protein supplementation. Cows fed protein supplements partitioned less energy toward body weight gain, but tended to partition more energy toward milk production. Efficiency of use of feed N to milk N increased by feeding SRUT and YMPT, and milk N-to-manure N ratio increased with YMPT. Overall results from this experiment indicate that replacing the mixture of soybean meal and canola meal with SRU and YMP in alfalfa hay-based dairy diets can be a good approach to improve nutrient utilization efficiencies in lactating dairy cows.

This experiment was conducted to determine the effects of corn silage (CS) hybrids and quality of alfalfa hay (AH) in high-forage dairy diets on N utilization, ruminal fermentation, and lactational performance by early-lactating dairy cows. Eight multiparous Holstein cows were used in a duplicated 4 × 4 Latin square experiment with a 2 × 2 factorial arrangement of dietary treatments. The 8 cows (average days in milk = 23 ± 11.2) were surgically fitted with ruminal cannula, and the 2 squares were conducted simultaneously. Within square, cows were randomly assigned to a sequence of 4 diets: conventional CS (CCS) or brown midrib CS (BMR) was combined with fair-quality AH [FAH: 46.7% neutral detergent fiber (NDF) and 18.4% crude protein (CP)] or high-quality AH (HAH: 39.2% NDF and 20.7% CP) to form 4 treatments: CCS with FAH, CCS with HAH, BMR with FAH, and BMR with HAH. Diets were isonitrogenous across treatments, averaging 15.9% CP. Each period lasted a total of 21 d, with 14 d for treatment adaptation and 7d for data collection and sampling. Intake of DM and milk yield did not differ in response to CS hybrids or AH quality. Although feeding BMR-based diets decreased urinary N output by 24%, it did not affect fecal N output. Feeding HAH decreased urinary N output by 15% but increased fecal N output by 20%. Nitrogen efficiency [milk N (g/d)/intake N (g/d)] tended to increase for BMR treatments. Ruminal ammonia-N concentration was lower for cows fed BMR-based diets than for those fed CCS-based diets but was not affected by quality of AH. Feeding BMR-based diets or HAH decreased milk urea N concentration by 23 or 15%, respectively, compared with CCS-based diets or FAH. Total volatile fatty acid concentration increased with HAH but was not influenced by CS hybrids. Feeding BMR-based diets decreased urinary N-to-fecal N ratio (UN:FN), and it was further reduced by feeding HAH. Although cows fed the BMR-based diets tended to increase milk N-to-manure N ratio, the quality of AH did not affect the ratio. The lower ratio of UN:FN with a higher ratio of milk N-to-manure N ratio for the BMR-based diets indicates that feeding BMR may reduce manure ammonia-N by reducing excretion of urinary N and increasing secretion of milk N per unit of manure N excreted.

This experiment was conducted to test a hypothesis that lactating dairy cows fed 35% brown midrib (BMR) corn silage and 25% alfalfa hay (dry matter (DM) basis) would consume more DM around peak lactation compared with those fed conventional corn silage (CS), resulting in longer peak milk production. Twenty-eight multiparous Holstein cows were used starting at the onset of lactation through 180 d in milk (DIM). Treatments were formulated to maintain a forage-to-concentrate ratio of 60:40, differing only in the CS hybrids used. Two dietary treatments were assessed in a completely randomized design: total mixed ration based on conventional CS (CCS) and total mixed ration based on BMR silage. Through peak lactation (1-60 DIM), DM intake was not different between dietary treatments, whereas DM intake post-peak lactation (61-180 DIM) tended to increase by feeding the BMR diet compared with the CCS diet (25.8 vs. 24.7 kg/d). Cows fed the BMR diet tended to lose less body weight through peak lactation compared with those fed the CCS diet (-0.22 vs. -0.52 kg/d). Although milk yield was not different between dietary treatments through peak lactation, milk yield post-peak lactation increased by feeding the BMR diet compared with the CCS diet (41.0 vs. 38.8 kg/d). Yield of 3.5% fat-corrected milk was similar between dietary treatments throughout the experiment (41.4 kg/d, on average), but milk fat concentration decreased by feeding the BMR diet compared with the CCS diet post-peak lactation (3.47 vs. 3.80%). Overall milk protein concentration was similar between dietary treatments throughout the experiment (2.96%, on average), whereas milk protein yield tended to be higher for the BMR diet post-peak lactation compared with the CCS diet (1.19 vs.1.13 kg/d). Feeding BMR silage with a high dietary concentration of alfalfa hay maintained more body weight, but did not affect milk production through peak lactation; however, cows fed the BMR diet post-peak lactation consumed more feed and maintained longer peak milk yield, leading to greater overall milk production and milk protein yield.

Twenty-two Kiko crossbred male goats (Capra hircus; initial BW = 27.5 ± 1.04 kg) were used in a randomized complete block design to determine the effects of feeding pine bark (PB; Pinus taeda L.) on animal performance, rumen fermentation, blood parameters, fecal egg counts (FEC), and carcass characteristics in goats. Experimental treatments included the control diet [0% PB plus 30% wheat straw (WS)], 15% PB plus 15% WS, and 30% PB plus 0% WS (on as-fed basis), where PB replaced WS. Freshly air-dried PB and WS were finely (1.5 to 3.0 mm) ground and incorporated in the grain mixes. Experimental diets provided a total of 1.9, 16.3, and 32 g of condense tannins (CT)/kg DM in 0%, 15%, and 30% PB diets, respectively. The grain mixes were fed daily at 85% of the feed offered, with remaining 15% consisting of Bermuda grass hay (Cynodon dactylon). Animals were fed once a day at 0800 h, and feed offered and refused was monitored for an 83-d performance period. Rumen and blood samples were collected at d 0, 50, and 80 of the study. Carcass traits were assessed after slaughter at the end of performance period. There was no difference in initial BW, hay, and total NDF intake among treatments; however, final BW (P = 0.06), ADG (P < 0.01), grain mix intake (P < 0.001), total DMI (P < 0.001), and G:F (P < 0.04) increased linearly as the PB increased in the diets. Rumen ammonia N, acetate, isovalerate and acetate-to-propionate ratio were reduced linearly (P < 0.05). There was no difference in carcass traits except cold carcass weight (P = 0.06), which tended to increase linearly in goats fed 15% and 30% PB. Breast, sirloin, trim trait, liver, and hide weight increased (linear; P < 0.01) with addition of PB. Blood basophils, alanine transaminase, aspartate aminotransferase, albumin, Na, and Cl concentrations decreased (linear; P < 0.02 to 0.01) as PB supplementation increased. Supplementation of PB reduced (linear; P < 0.01) average FEC. Addition of PB in the diets improved performance, reduced FEC, and favorably modified rumen fermentation.

A lactation trial was conducted to determine the effects of supplementing whole safflower seeds (SS) on ruminal fermentation, lactational performance, and milk fatty acid (FA) profiles. Nine multiparous Holstein cows (days in milk = 110 ± 20) were used in a replicated 3 × 3 Latin square design. Each period lasted 21 d, with 14 d of adaptation and 7 d of data collection. Within square, cows were randomly assigned to a sequence of 3 dietary treatments as follows: cottonseed total mixed ration (TMR; CST), conventional SS (variety S-208) TMR (CSST), and NutraSaff SS (Safflower Technologies International, Sidney, MT) TMR (NSST). Diets contained approximately 63% forage (36% alfalfa hay, 4% grass hay, and 23% corn silage) and 37% concentrate supplemented with 2% cottonseed to the CST and 3% conventional or NutraSaff SS to the CSST or the NSST, respectively. Intake of dry matter (DM) averaged 21.8 kg/d and did not differ across diets, but feeding the NSST decreased intake of neutral detergent fiber (NDF) due to lower dietary concentration of NDF in the NSST. Digestibilities of DM and nutrients were similar among treatments. No differences in yields of milk or milk components were observed in response to supplementing SS. Dietary treatments did not affect ruminal pH, total or molar proportions of ruminal volatile FA, and ammonia-N. However, cows fed SS had a higher molar proportion of isobutyrate than those fed the CST diet. Ruminal C16:0 FA concentration increased with the CST, whereas C18:1 cis-9 and C18:2 n-6 tended to increase with SS supplementation, indicating that conventional and NutraSaff SS were partially protected from microbial biohydrogenation. Supplementing SS decreased milk C16:0 concentration, whereas it increased C18:1 cis-9 and C18:1 trans-9. Milk FA C18:1 trans-11 and cis-9, trans-11 conjugated linoleic acid increased and tended to increase with feeding the NSST, respectively, but not the CSST diet. In conclusion, supplementing diets with whole SS at 3% of dietary DM can be an effective strategy of fat supplementation to lactating dairy cows without negative effects on lactational performance and milk FA profiles.

A lactation experiment was conducted to determine the influence of quebracho condensed tannin extract (CTE) on ruminal fermentation and lactational performance of dairy cows. The cows were fed a high forage (HF) or a low forage (LF) diet with a forage-to-concentrate ratio of 59:41 or 41:59 on a dry matter (DM) basis, respectively. Eight multiparous lactating Holstein cows (62 ± 8.8 d in milk) were used. The design of the experiment was a double 4 × 4 Latin square with a 2 × 2 factorial arrangement of treatments, and each period lasted 21 d (14 d of treatment adaptation and 7 d of data collection and sampling). Four dietary treatments were tested: HF without CTE, HF with CTE (HF+CTE), LF without CTE, and LF with CTE (LF+CTE). Commercial quebracho CTE was added to the HF+CTE and the LF+CTE at a rate of 3% of dietary DM. Intake of DM averaged 26.7 kg/d across treatments, and supplementing CTE decreased intakes of DM and nutrients regardless of forage level. Digestibilities of DM and nutrients were not affected by CTE supplementation. Milk yield averaged 35.3 kg/d across treatments, and yields of milk and milk component were not influenced by CTE supplementation. Negative effects of CTE supplementation on feed intake resulted in increased feed efficiency (milk yield/DM intake). Although concentration of milk urea N (MUN) decreased by supplementing CTE in the diets, efficiency of N use for milk N was not affected by CTE supplementation. Feeding the LF diet decreased ruminal pH (mean of 6.47 and 6.33 in HF and LF, respectively). However, supplementation of CTE in the diets did not influence ruminal pH. Supplementing CTE decreased total volatile fatty acid concentration regardless of level of forage. With CTE supplementation, molar proportions of acetate, propionate, and butyrate increased in the HF diet, but not in the LF diet, resulting in interactions between forage level and CTE supplementation. Concentration of ammonia-N tended to decrease with supplementation of CTE. The most remarkable finding in this study was that cows fed CTE-supplemented diets had decreased ruminal ammonia-N and MUN concentrations, indicating that less ruminal N was lost as ammonia because of decreased degradation of crude protein by rumen microorganisms in response to CTE supplementation. Therefore, supplementation of CTE in lactation dairy diets may change the route of N excretion, having less excretion into urine but more into feces, as it had no effect on N utilization efficiency for milk production.

This experiment was conducted to determine the effects of corn silage hybrids and nonforage fiber sources (NFFS) in high forage diets formulated with high dietary proportions of alfalfa hay (AH) and corn silage (CS) on ruminal fermentation and productive performance by early lactating dairy cows. Eight multiparous Holstein cows (4 ruminally fistulated) averaging 36±6.2 d in milk were used in a duplicated 4 × 4 Latin square design experiment with a 2 × 2 factorial arrangement of treatments. Cows were fed 1 of 4 dietary treatments during each of the four 21-d replicates. Treatments were (1) conventional CS (CCS)-based diet without NFFS, (2) CCS-based diet with NFFS, (3) brown midrib CS (BMRCS)-based diet without NFFS, and (4) BMRCS-based diet with NFFS. Diets were isonitrogenous and isocaloric. Sources of NFFS consisted of ground soyhulls and pelleted beet pulp to replace a portion of AH and CS in the diets. In vitro 30-h neutral detergent fiber (NDF) degradability was greater for BMRCS than for CCS (42.3 vs. 31.2%). Neither CS hybrids nor NFFS affected intake of dry matter (DM) and nutrients. Digestibility of N, NDF, and acid detergent fiber tended to be greater for cows consuming CCS-based diets. Milk yield was not influenced by CS hybrids and NFFS. However, a tendency for an interaction between CS hybrids and NFFS occurred, with increased milk yield due to feeding NFFS with the BMRCS-based diet. Yields of milk fat and 3.5% fat-corrected milk decreased when feeding the BMRCS-based diet, and a tendency existed for an interaction between CS hybrids and NFFS because milk fat concentration further decreased by feeding NFFS with BMRCS-based diet. Although feed efficiency (milk/DM intake) was not affected by CS hybrids and NFFS, an interaction was found between CS hybrids and NFFS because feed efficiency increased when NFFS was fed only with BMRCS-based diet. Total volatile fatty acid production and individual molar proportions were not affected by diets. Dietary treatments did not influence ruminal pH profiles, except that duration (h/d) of pH <5.8 decreased when NFFS was fed in a CCS-based diet but not in a BMRCS-based diet, causing a tendency for an interaction between CS hybrids and NFFS. Overall measurements in our study reveal that high forage NDF concentration (20% DM on average) may eliminate potentially positive effects of BMRCS. In the high forage diets, NFFS exerted limited effects on productive performance when they replaced AH and CS. Although the high quality AH provided adequate NDF (38.3% DM) for optimal rumen fermentative function, the low NDF concentration of the AH and the overall forage particle size reduced physically effective fiber and milk fat concentration.

A series of in vitro fermentation experiments was performed to assess the effects of 4 feed enzyme products (FE) that varied in enzymatic activities on the degradation of alfalfa hay and corn silage. The FE contained a range of endoglucanase, exoglucanase, xylanase, and protease activities, and a range of dose rates (DR) was used. The objective of the study was to identify effective formulations and optimum DR, and to establish if combining FE would further improve fiber degradation. For alfalfa hay, quadratic increases in gas production and degradation of dry matter (DM) and fiber were observed for all FE, with maximum responses at low to medium DR. For corn silage, none of the FE increased gas production or DM degradation, but all FE increased NDF degradation, with optimum DR in the low to medium range. The proteolytic enzyme papain improved fiber degradation of alfalfa hay and corn silage in a manner similar to that observed for polysaccharidase FE. Among the polysaccharidase FE, added activities of endoglucanase and exoglucanase were positively correlated with improvement in neutral detergent fiber (NDF) degradability of corn silage, whereas only added endoglucanase activity tended to be correlated with improvement in NDF degradability of alfalfa hay. Combining effective polysaccharidase FE further improved fiber degradation of both forages, with greater improvements for corn silage. Combining polysaccharidase and proteolytic FE further improved NDF degradation of corn silage, but not alfalfa hay. Combination treatments generally resulted in additive effects with increases in fiber degradation equal to the sum of the improvements for the individual enzyme components. Improved fiber degradation of corn silage was associated with decreased acetate to propionate ratios. Enzyme products that improve in vitro degradation of forages may have the potential to improve lactational performance of dairy cows.

Two in vitro experiments were performed to identify promising exogenous fibrolytic enzyme products (EFE) and optimum dose rates (DR) for improving the degradation of alfalfa hay and corn silage. The relationship between enzymatic activity and fermentation responses was examined to identify optimum formulations. In experiment 1, 5 EFE containing mainly endoglucanase and xylanase activities, with different ratios between the 2 activities, were assessed at a DR of 0.7, 1.4, and 2.1 mg/g of DM forage. Milled alfalfa hay or corn silage was incubated in an in vitro batch culture with buffer, ruminal fluid, and EFE. Gas production (GP) was measured during 24 h of incubation, and degradabilities of DM and fiber were measured after terminating the incubation at 24 h. Two (E1 and E3) EFE substantially improved GP and degradation of alfalfa hay and corn silage fiber. The optimum DR of these EFE was 1.4 mg/g of DM for both forages with improvements in NDF degradability up to 20.6% for alfalfa hay and up to 60.3% for corn silage. Whereas added activities of endoglucanase and exoglucanase were positively correlated with improvement in NDF degradability for alfalfa hay and corn silage, there was no relationship between added xylanase activity and NDF degradability. The 2 most promising EFE from experiment 1 were reevaluated in experiment 2, alone and in combination with a high xylanase EFE, to determine whether their effectiveness could be enhanced by decreasing the endoglucanase to xylanase ratio. The 2 EFE improved GP and fiber degradation in a manner similar to that observed in experiment 1, but the combination treatments resulted in no further beneficial effects. Exogenous fibrolytic enzyme products can greatly improve forage utilization, but DR and the activities supplied are critical for achieving this response. Products used with alfalfa hay and corn silage should contain high endoglucanase activity, with an ideal ratio of endoglucanse to xylanase.

PURPOSE: To evaluate the clinical outcome of the extrapedicular approach of percutaneous vertebroplasty (PVP) for upper and mid-thoracic vertebral compression fractures in patients. MATERIAL AND METHODS: Extrapedicular vertebroplasty was performed in painful compression fractures at T4-T8 levels. The assessment criteria were changes over time in visual analog scale (VAS) and mobility score. We evaluated the volume of cement injected, the size of needle required, and complications. RESULTS: Procedures were performed in 27 patients with a total of 34 affected vertebral bodies. Early (within a week) and one year later, clinical follow-ups showed that pain intensity had decreased by 50% one day after operation and later by 70-80%. Mobility scores of all patients were improved immediately after the procedure. Average volume of polymethylmethacrylate (PMMA) per vertebral body was 3.8 +/- 1.2 ml. Leakage of PMMA occurred in one vertebral level (intradiskal space), but did not cause clinical complications. CONCLUSION: PVP of upper and mid-thoracic spine with an extrapedicular approach is an efficient and safe procedure for treating painful thoracic vertebral compression fracture under a cautious patient selection and meticulous technical procedure.

The effects of exogenous proteolytic enzyme (EPE) on intake, digestibility, ruminal fermentation, and lactational performance were determined using 8 lactating Holstein cows in a double 4 x4 Latin square experiment with a 2 x2 factorial arrangement of treatments. Diets based on barley silage and alfalfa hay as the forage sources were formulated to maintain different forage to concentrate ratios [60:40 vs. 34:66, dry matter (DM) basis]. Four dietary treatments were tested: high forage (HF) without EPE (HF-EPE), HF with EPE (HF+EPE), low forage (LF) without EPE (LF-EPE), and LF with EPE (LF+EPE). The EPE, which contained proteolytic activity but negligible fibrolytic activity, was added to the concentrate portion of the diets after pelleting at a rate of 1.25 mL/kg of DM. Adding EPE to the diet increased total tract digestibilities of DM, organic matter, N, acid detergent fiber, and neutral detergent fiber, with larger increases in digestibility observed for cows fed LF+EPE. Effects of added EPE on in vivo digestibility were consistent with improvements in gas production and degradability of the individual components of the TMR observed in vitro. Ruminal enzymic activities of xylanase and endoglucanase increased with addition of EPE to the diet, which may have accounted for improvements in fiber digestion. However, feeding EPE unexpectedly decreased feed intake of cows, which offset the benefits of improved feed digestibility. Consequently, milk yield of cows fed high or low forage diets decreased with adding EPE. Nevertheless, dairy efficiency, expressed as milk/DM intake, was highest for the LF+EPE diet. Addition of EPE to the diet increased milk fat and milk lactose percentages, but decreased milk protein percentage of cows fed a low forage diet. For cows fed high forage diets, EPE only increased milk lactose percentage. Efficiency of N use for milk production was decreased for both the high and low forage diets when EPE was added to the diet. Mean ruminal pH was lowered when EPE was added a low forage diet, likely due to the increased degradation of forage and concentrate, but there was no effect of EPE on rumen pH when cows were fed high forage diets. Profiles of VFA and microbial yield were not affected by adding EPE to the diets. Adding EPE to a total mixed ration containing alfalfa hay, barley silage, and concentrate improved nutrient digestibility in the total tract, and the response was maximized with a high concentrate diet. However, improvements in digestibility were offset by decreased feed intake, likely due to increased ruminal acidosis.

Mechanical processing of whole crop barley before ensiling may be useful for improving nutrient use by dairy cattle. The objective of this study was to assess the effects of feeding mechanically processed barley silage as the main forage source on lactational performance. Twenty-four Holstein cows, 16 primiparous (187 +/- 52 days in milk) and 8 multiparous (87 +/- 69 days in milk) cows, were used in a completely randomized design with a 2-wk covariate period and a 6-wk treatment period. The 2 treatments were: 1) total mixed ration (TMR) containing regular barley silage (RBS-TMR), and 2) TMR containing mechanically processed barley silage (MPBS-TMR). Barley silage and alfalfa hay supplied 41 and 5% of the dietary dry matter (DM), respectively. Intake, body weight, and milk production were measured during the covariate and treatment periods. In addition, 2 multiparous cows were used for in situ measurements of the ruminal DM and fiber degradation kinetics of the barley silages and TMR. Data were analyzed with repeated measurements using a mixed model that included the covariate adjustment. Feeding MPBS-TMR had no significant effects on DM intake (DMI; 21.7 kg/d), milk yield (33.9 kg/d), or milk composition, with only 4% FCM (fat-corrected milk) yield (29.7 vs. 31.7 kg/d) and milk fat concentration (3.30 vs. 3.57%) showing a numerical improvement. Apparent digestibilities of DM and nutrients were not affected by feeding MPBS-TMR, with the exception of starch digestibility, which tended to increase. Dairy efficiencies calculated as milk yield/DMI or FCM/DMI were not different between treatments. Body weight and body condition score were not affected by treatments. Effective ruminal degradability of DM was similar for both barley silages, indicating that when the silages were ground to remove the effects of mechanical processing, the potential digestion was similar. Mechanical processing of barley silage harvested at a mid-dough stage of maturity resulted in small improvements in its nutritive value for lactating dairy cows and had minor impact on digestibility and milk production.

Five dual-flow fermentors (700 mL) were used to determine the effects of eastern gamagrass (Tripsacum dactyloides [L.] L.) diets on microbial metabolism by mixed rumen cultures. Fermentors were incubated with filtered ruminal contents and allowed to adapt for 4 d to diets followed by 3 d of sample collection. Five dietary treatments were tested: 1) gamagrass hay (GH) + no corn (GHNC), 2) gama grass silage (GS) + no corn (GSNC), 3) GS + low corn (GSLC), 4) GS + medium corn (GSMC); and 5) GS + high corn (GSHC). The experiment was conducted as a randomized complete block design with five treatments and three replications. Total VFA concentrations were not affected by diets. Corn addition linearly decreased (P < 0.001) molar proportion of acetate. In contrast, molar proportion of propionate was reduced in GSLC (cubic effect, P < 0.001) but remained similar across other diets. Corn supplementation linearly increased molar proportion of butyrate (P < 0.001). The acetate + butyrate-to-propionate ratio was highest in cultures offered GSLC (cubic effect, P < 0.001) but similar across other diets. Feeding GSNC resulted in a higher ruminal pH compared with GHNC (P < 0.03). Increasing the level of corn supplementation in GS linearly decreased culture pH (P < 0.001). All diets resulted in similar methane production, with the exception of GSMC, which lowered methane output (quadratic effect, P < 0.004). Total substrate fermented to VFA and gas tended to be greater with GHNC than with GSNC (P < 0.06) and linearly increased with the addition of corn (P < 0.004). Neutral detergent fiber digestibility was similar between GH and GS and was not affected by supplemental corn. Microbial N flow increased in cultures offered GSHC (quadratic effect, P < 0.02). Corn supplementation at the medium and high level linearly decreased C 18:0 (P < 0.02) and increased trans-C18:1 (P < 0.004). Including corn at the high level with GS did not have a detrimental effect on fermentation in dual-flow fermentors.
